FORT LAUDERDALE, FL -- October 25, 1999 -- New research shows topical civamide is effective in reducing pain associated with primary, post-traumatic osteoarthritis of the knee.

The researchers were Joel E. Bernstein and Scott B. Phillips, of Winston Laboratories; Rashid Khairi, of Physicians Research Group; and Charles Birbara, of Clinical Pharmacology Study Group. The researchers studied civamide (cis-8-methyl-N-vanilly1-6-nonenamide), a vanilloid receptor agonist and neuronal calcium channel blocker that inhibits the release of excitatory neurotransmitters and depletes neurons of their neuropeptide contect.

In a double-blind, vehicle-controlled, parallel study, researchers recruited 179 patients (at least 40 years of age) with moderate to severe, primary or post-traumatic osteoarthritis (OA) of the knee. They applied either 0.075 percent civamide cream or cream vehicle QID to the knees for four weeks.

The results show that, after seven days of treatment, civamide-treated patients had nearly a 40 percent reduction in pain, evaluated by the visual analogue scale (VAS). After four weeks of treatment, pain was reduced by a mean of 55 percent, with statistically significant reductions in knee pain observed for civamide treated joints versus those treated with vehicle for all four efficacy variables measured.

The investigators reported that the only side effect associated with civamide was transient burning and stinging reactions at the application sites. They concluded that civamide 0.075 percent cream is a safe and effective treatment for pain associated with OA of the knees.
