Temozolomide is can be used as a second-line regimen in patients heavily pre-treated for recurrent glioblastoma multiforme. Italian researchers report progression-free survival with temozolomide was 24 percent at six months in patients with recurrent brain tumours. Progression-free survival at 12 months was 8 percent. Investigators from Azienda Ospedale-Universita sites at both Padova and Verona conducted a phase II study with 42 glioblastoma patients to assess the efficacy of the drug. All of the participants had had surgery plus radiotherapy and a first-line regimen based on nitrosourea, procarbazine and vincristine. They received 150 milligrams/m[² of temozolomide daily for five days every four weeks. Response in all of the participants was assessed and median time to progression was found to be 11.7 weeks. Overall, complete response was observed in two patients (4.7 percent) and partial response in six patients (14.3 percent). Disease was stabilised in nine patients (21.4 percent). The researchers concluded: "Temozolomide as a second-line regimen is a valid option in patients with heavily pre-treated glioblastoma multiforme."
