Pregabalin Appears Efficacious for Treatment Across Spectrum of Generalised Anxiety Disorder Subtypes

MIAMI, FL -- March 16, 2004 -- Pregabalin appears to be efficacious for treatment of patients with various subtypes of generalized anxiety disorder (GAD), investigators reported here on March 13th at the Anxiety Disorders Association of America 24th Annual Conference.

To evaluate the efficacy of pregabalin within "clinically relevant subtypes" of GAD, the researchers collected data from placebo-controlled phase 2 and 3 studies in which pregabalin was investigated as a treatment option for individuals who met Diagnostic and Statistical Manual ? Revision IV criteria for GAD.

They combined all pregabalin dosage groups for a total of 1,282 patients, who were given dosages ranging from 200 to 600 mg/day. The authors stated that all subgroups were analysed according to gender, age, disease severity determined by the 15-question Hamilton Anxiety Rating Scale (HAM-A 15), severe somatic symptoms (HAM-A somatic-subsyndromic depression (HAM-D 12), and severe insomnia. They then performed a week 4/6 last observation carried forward (LOCF) end point analysis with responders defined as those having a 50% reduction in HAM-A total score.

Dr. Pollack and his team found that rates of response were significantly greater among in the pregabalin group than those in the placebo group among men (56% vs. 39%; P < .001), among women (51% vs. 32%; P < .001), among the elderly (55% vs. 27%; P < .02) and among patients with severe anxiety (61% vs. 39%; P < .001), subsyndromic depression (49% vs. 31%; P < .001), severe somatic symptoms (57% vs. 35%; P < .0001), or severe insomnia (60% vs. 37%; P < .001).

In addition, investigators noted significantly greater improvement in symptom factors used to define high-severity subgroups, including HAM-A somatic factor (week 4/6 LOCF end point change score ?7.3 vs. ?5.3; P < .0001).

Pregabalin is efficacious across the spectrum of clinically relevant subtypes of patients with GAD, the study authors concluded.
